Stem cells and their malignant counterparts require the support of a specific microenvironment or “niche”. While various anti‐cancer therapies have been broadly successful, there are growing opportunities to target the environment in which these cells reside to further improve therapeutic efficacy and outcome. This is particularly true when the aim is to target normal or malignant stem cells. The field aiming to target or use the niches that harbor, protect, and support stem cells could be designated as “nichotherapy”. In this (...) essay, we provide a few examples of nichotherapies. Some have been employed for decades, such as hematopoietic stem cell mobilization, whereas others are emerging, such as chemosensitization of leukemia stem cells by targeting their niche. (shrink)

Is inheritable genetic modification the new dividing line in gene therapy? The editors of this searching investigation, representing clinical medicine, public health and biomedical ethics, have established a distinguished team of scientists and scholars to address the issues from the perspectives of biological and social science, law and ethics, including an intriguing Foreword from Peter Singer. Their purpose is to consider how society might deal with the ethical concerns raised by inheritable genetic modification, and to re-examine prevailing views about whether (...) these procedures will ever be ethically and socially justifiable. The book also provides background to define the field, and discusses the biological and technological potential for inheritable genetic modification, its limitations, and its connection with gene therapy, cloning, and other reproductive interventions. Until recently, retention of introns in mature mRNAs has been regarded as a consequence of mis‐splicing. Intron‐retaining transcripts are thought to be non‐functional because they are readily degraded by nonsense‐mediated decay. However, recent advances in next‐generation sequencing technologies have enabled the detection of numerous transcripts that retain introns. As we review herein, intron‐retaining mRNAs play an essential conserved role in normal physiology and an emergent role in diverse diseases. Intron retention should no longer be overlooked as a key mechanism that (...) independently reduces gene expression in normal biology. John Woods' The Logic of Fiction, now thirty-five years old, is a ground-breaking event in the establishment of the semantics of fiction as a stand-alone research programme in the philosophies of language and logic. There is now a large literature about these matters, but Woods' book retains a striking freshness, and still serves as a convincing template of the treatment options for the field's key problems. The book now appears in a second edition with a new Foreword by Nicholas (...) Griffin and an extended bibliography covering the period 1969-2009. As Griffin notes in his Foreword, it is "surprising on looking back to discover how little was written on the semantics of fiction before John Woods' The Logic of Fiction was published in 1974. Nowadays, science is treated an instrument of policy, serving the material interests of government and commerce. Traditionally, however, it also has important non-instrumental social functions, such as the creation of critical scenarios and world pictures, the stimulation of rational attitudes, and the production of enlightened practitioners and independent experts. The transition from academic to ‘post-academic’ science threatens the performance of these functions, which are inconsistent with strictly instrumental modes of knowledge production. In particular, expert objectivity is negated by entanglement with (...) political and commercial interests. We cannot go back to the old academic model for science, but need to consider how to maintain its vital non-instrumental roles. (shrink)

Abstract This paper attempts to clarify the debate between those philosophers who hold that the development of science is governed by objective standards of rationality and those sociologists of science who deny this. In particular it focuses on the debate over the ?symmetry thesis?. Bloor and Barnes argue that a properly scientific approach to science itself demands that an investigator should seek the same general type of explanation for all decisions and actions by past scientists, quite independently of whether or (...) not she or he happens to agree with those decisions or approve those actions as ?correct? or ?rational?. I try to improve on previous treatments of the ?rationalist? position (by Lakatos, Laudan, Newton?Smith and Brown) and clarify the exact asymmetries to which the ?rationalist? is, and is not, committed. This second set of papers by John Rist is concerned with attempts by (mostly pagan) thinkers in Greco-Roman antiquity to understand the nature of morality against a background of wide-ranging debate about the relationship between soul and body and the necessity for a correct psychology and physiology if the 'good life for man' is to be revealed. Three papers are on Plato, whose elaborate mix of ethics, psychology and metaphysics sets the stage for most of the debate; one is (...) on Aristotle, five are on the Stoics and five on Plotinus. A further study deals with the general problem of the relationship between ethics, cosmology and biology, and the series concludes with the crisis among both pagans and Christians in late antiquity over whether man is naturally good enough to correct his own moral weakness. Richard Rowland has recently argued that considerations based on moral disagreement between epistemic peers give us reason to think that cognitivism about moral judgments, i.e., the thesis that moral judgments are beliefs, is false. The novelty of Rowland’s argument is to tweak the problem descriptively, i.e., not focusing on what one ought to do, but on what disputants actually do in the light of peer disagreement. The basic idea is that moral peer disagreement is intelligible. However, if moral judgments were (...) beliefs, and beliefs track perceived evidence, then moral peer disagreement would not be intelligible. Hence, moral judgments are not beliefs. The argument is both novel and interesting, but this paper argues that it fails to establish the conclusion. Beliefs are plausibly analyzed as constituted by dispositions to respond to what is perceived as evidence, but dispositions can always be interfered with. Provided a background explanation of why the disposition is not manifested, peer intransigence is quite intelligible. (shrink)

Western thought since the seventeenth century has been dominated by methodological solipsism (Krieger, 1991). The famous sound-bite of René Descartes 'cogito, ergo sum': 'I think, therefore I am', became the starting point for most discourse on the nature of things. This dictum does not advocate idealism. It does not assert that everything is necessarily a construct of the human mind. But it assumes that the world of things and beings is surveyed and interpreted from the point of view of a (...) single individual. Nowhere is this stance more entrenched than in the philosophy of science. To a remarkable degree the scientist is represented as studying the natural world as if alone in it, served only by mindless assistants who might as well be replaced by machines. Scientific theories are presented as systems of thought conjured up and tested by that same individual in a further series of single-headed operations. Research results are formulated and treated philosophically as the independent findings of lone explorers, each reporting the evidence of their own eyes and their rational inferences concerning the hidden mechanisms by which these personal percepts might be generated. Our epistemological role models are Robinson Crusoe and Sherlock Holmes, self-sufficient intellectuals to whom their human companions, Friday and Watson, are mere stooges. (shrink)
